Multiple Feedback Loops in Circadian Cycles: Robustness and Entrainment as Selection Criteria

چکیده
This paper discusses the contribution of an additional feedback loop to the entrainment and the robustness of a circadian system. To quantify robustness, we perform a global analysis of the system’s parameter space. We quantify the parameter region where the circuit displays an experimentally observed behavior, under entrainment. This global measure is comleted with a classification based on the phase response curve (PRC). For two models of circadian rhythms, we found that the one with two loops is more robust than the one with a single loop: the two-loop model shows better resilience to parameter perturbations and it has also a larger region where the PRC matches experimental PRCs of circadian oscillators.

A Symmetric Dual Feedback System Provides a Robust and Entrainable Oscillator
Many organisms have evolved molecular clocks to anticipate daily changes in their environment. The molecular mechanisms by which the circadian clock network produces sustained cycles have extensively been studied and transcriptional-translational feedback loops are common structures to many organisms. Although a simple or single feedback loop is sufficient for sustained oscillations, circadian ...
متن کاملWeather and Seasons Together Demand Complex Biological Clocks
The 24-hour rhythms of the circadian clock [1] allow an organism to anticipate daily environmental cycles, giving it a competitive advantage [2, 3]. Although clock components show little protein sequence homology across phyla, multiple feedback loops and light inputs are universal features of clock networks [4, 5]. Why have circadian systems evolved such a complex structure? All biological cloc...
متن کاملDiscrete Delay Model for the Mammalian Circadian Clock
A three-variable discrete delay model is proposed for the circadian rhythm of the mammals with BMAL1, PER-CRY complex and REV-ERB protein concentrations as the dynamical variables. The delay model is phenomenological in nature rather than the precise description of all the underlying complex processes. The goal of this paper is to study the effects of delay in the circadian rhythms of mammals t...
متن کاملCellular Circadian Pacemaking and the Role of Cytosolic Rhythms
The daily rhythms that adapt organisms to the solar cycle are driven by internal circadian clocks. The hypothesis that the core pacemakers of these clocks consist of auto-regulatory transcriptional/post-translational feedback loops (TTFLs) was first developed in flies and fungi and has now been extended successfully to describe circadian timing mechanisms in mammals and plants. TTFL models revo...
متن کاملA reduced model clarifies the role of feedback loops and time delays in the Drosophila circadian oscillator.
Although several detailed models of molecular processes essential for circadian oscillations have been developed, their complexity makes intuitive understanding of the oscillation mechanism difficult. The goal of the present study was to reduce a previously developed, detailed model to a minimal representation of the transcriptional regulation essential for circadian rhythmicity in Drosophila. ...
